Abstract
While silent in normal differentiated human tissues, telomerase is reactivated in most human cancers. Thus, telomerase is an almost universal oncology target. This update describes preclinical and clinical advancements using a variety of approaches to target telomerase. These include direct telomerase inhibitors, G-quadruplex DNA-interacting ligands, telomerase-based vaccine platforms, telomerase promoter-driven attenuated viruses, and telomerase-mediated telomere targeting approaches. While imetelstat has been recently approved by the Food and Drug Administration (FDA), several other approaches are in late-stage clinical development. The pros and cons of the major approaches will be reviewed.

Preclinical and Clinical advancements are always exciting, but I’d say more exciting in the rear view mirror. I highly doubt any one investing in Geron or reading this board is pumped by potential advancements that may come in the upcoming years or decades.
I can barely wait another few weeks for European marketing authorization let alone years or decade(s). I’m struggling over here and I see the Institutions who now control the vast amount of the stock are (trying to) prey on that obvious retail / small fry sentiment. Not going to happen over here despite my beyond frayed nerves.
(And yes it is a correct notion that an inept and feckless, not believing in science administration leading the NIH, FDA etc will be a further setback if not worse… not sure why that such an opinion or statement should be forbidden when Super Dave, curb your enthusiasm ((a personal favorite)), and rock stars / music videos are so prolific. I say bring it all and people could read and respond to what they want.)

Bp - correct, that was actually mentioned yesterday in his "don't screw us thread"......

As far as AI is concerned, not exactly a reliable source of information. It misses quite a bit. Perhaps a decent starting point if you feel the need, but there are other therapies targeting the same indications as Imetelstat, which are gaining traction in CT's.
I think one of Imetelstats most unique qualities will be in its abilities as a combination option. If you take a look at Keytruda (pembrolizumab) by Merck, about 2/3rds of its FDA approvals (there are 41 now) are in combination with other drugs - including ruxolitinib. This was a drug that was almost overlooked in its infancy and shelved. As a matter of fact, when it was in the hands of Schering Plough:
"Just when it seemed like the team was starting to gain momentum, Schering itself was acquired by Merck in 2009, initiating, of course, a new reprioritization process for the integrated companies. This time, the PD1 program was less fortunate, and the team was told to stop work. Yet, they persisted–in skunkworks fashion, variously pitching their work as an oncology project, an antiviral project and a vaccine enhancement project. Eventually, however, management caught on and formally shut down their work. The molecule was placed on the out-license list, and ultimately a term sheet was drafted setting the terms for a specific interested party to obtain rights to the molecule at what was said to be a negligible cost.
Everything changed, however, by the middle of 2010. First, BMS published in the New England Journal of Medicine (NEJM) a successful Phase III study in refractory metastatic melanoma involving ipilimumab (Yervoy), a drug targeting the different but related T-cell inhibitor molecule CTLA4, pointing to the potential efficacy of the so-called “checkpoint inhibitor” approach. "This was a well-tolerated combination with no DLTs in patients with intermediate-1, intermediate-2, or high-risk myelofibrosis who were already on ruxolitinib,” Mascarenhas said in conclusion. “The 2 drugs in combination did not affect each other’s pharmacokinetic profiles, and there is some early evidence of efficacy in terms of spleen, symptom, and [possibly] mutational changes.”

And any of those has zero political motivations unlike your above statement. There are numerous articles starting to suggest quite the opposite. And I will add that perhaps ceasing Big Pharma from political influence and the FDA from allowing toxins in our food, amongst other things might not only benefit mankind in the long run, but also investors such as ourselves who have undoubtedly suffered from the weight of both BP and the FDA the past 15 years or longer. -Kmall

What can legitimately be discussed here as related issues? What is now happening in public health, at NIH, and at the FDA both with cancer research and potential cancer vaccines is now fully in focus and it affects everyone. It is now part of our history. Unlike yourself, I haven't made ONE political statement on this board. Mentioning a Govt agency and employee of is not a POLITICAL statement. I simply mentioned her so that LWS was aware of who makes crucial decisions in that agency and the connections tied to that position. You seem to have a reading comprehension issue here.
No one is saying you can't have opposing views. Who stated that? Again, YOU are making something up.
The issue here are NON FACTUAL statements that are POLITICALLY motivated. Is that the purpose of this forum?
After Bp politely requested no politics on the board YOU threw this out yesterday:
"And yes it is a correct notion that an inept and feckless, not believing in science administration leading the NIH, FDA etc will be a further setback if not worse…"
Not only is the above statement completely unfounded, it's DISRESPECTFUL to the moderator after having asked to refrain from political views here.
A decent person would consider apologizing for continuing on with what is already on the verge of simply opinionated based hogwash.
Yes, try INCLUDING an actual link of a reliable source to back up claims. I believe that's part of the guidelines here. Otherwise these outlandish claims are MISINFORMATION. It helps to actually do your own homework Ryan.
And yes, I'll point something out if it's a load of rubbish, as I believe Bp does as well, although he is much more eloquent.
Now for the sake of this board it's time to give the Good Vibes thread a rest........it's nothing but BAD vibes lately. -Kmall

bp (my thoughts for your comment) -- It appears that we have reached an impasse here. This is your chatroom to monitor and control. If you allow one side to post an opinion, please let the other side respond without fear of retribution. We are in unprecedented times, where current events and recent history are interacting in ways that we have never seen before. How are truth and facts to be defined when so many are operating under the cover of "big lies"? Unfortunately, it seems that we have to put some practical limits on how we communicate with each other.

I think it is appropriate for LWS's concerns about the fate of the NIH and the FDA to be voiced on this forum. If other folks disagree with the facts he cites or his opinions they should feel free to express themselves as well.

Big difference between what is an actual fact and an opinion - and regardless, there is also a difference with a basic "concern" and a political statement.
Up until this point - with the exception of LWS and now Ryan yesterday, I have yet to read any other poster throw politics into the discussion here regarding any Govt agency.
If that's the way this board wants to continue, then I'll be more than happy to refrain from reading or participating here any longer. I just don't see how a political debate enhances this forum in any way, shape or form? -Kmall

I still believe that the vibes for Rytelo are good. If some of the items that I have posted seem to be political to some, it has been accidental. I apologize for that and will try to do better.
There seems to be a disagreement about what is political and what is not. I am very surprised at some of the comments about fact and opinion. As much as possible, I have documented what I have posted and have tried to separate facts from opinion. Sometimes there are disagreements about what are facts and what are not. Anything forward looking will have some element of opinion (possibilities & probabilities).
Here are some of the active subjects that are currently being discussed by me and others:
1. Rytelo (imetelstat) -- as is, new chemistry and new patents
2. Geron as a company -- earnings, leaders and possible partners or collaborators
3. Rytelo combinations
4. The FDA approvals
5. NIH and the FDA going forward
6. Pending approvals (EMA, EU, MHRA, etc.)
7. Rytelo as part of cancer vaccine research and as a supplement to chemotherapy
8. Blood cancer understanding as compared to other (solid) cancers
9. Potential competition and other approaches
10. KOLs, conferences, publications
I will use this list as a guide. I am prepared to be corrected when necessary, and do not take any criticism personally.
