Re: hayflicklim post on ymb about NCT01731951

Rob, I disagree, there has been nothing to suggest the study is not going well. The discontinuation of the low dose arm means nothing and the continuation of the high dose arm with suspension of enrollment could be looked at two ways. One, there is so much uncertainty that it is not worth the resour...

Re: hayflicklim post on ymb about NCT01731951

Rob, well technically I don't think you can conclude that patients are still alive in the pilot study. One of the study conclusion criterion (among others, including drug approval) is that no patients remain alive. We know from Irish that John has done spectacularly well and ligas3 has contributed i...

Re: hayflicklim post on ymb about NCT01731951

Rob, this study was updated several months ago. The updates reflect the presence of a DMB, a total of 81 patients in the study and a May analysis date. Its all there on the clinical trials site. This study is the Tefferi original pilot study from Mayo reassigned to Janssen. bp

CKTC post link

Fish, I found the repost from CKTC very thoughtful and was able only to access the link to the first article. Have you been able to link to the second one in Nature? I haven't. Please advise if there is another way to get there, thanks. bp

Alexander Fleming and Penicillin

I wonder if Dr. Rusbuldt et al felt a little like Alexander Flemming (who saw a ring of no bacterial growth in his petri dish because of, well you know the story)...when they saw multiple AML cells lines die after 96 hours...I know Cheng, petri dishes are not human, mice are not humans but this coul...

Re: Remark from CKTC on SA

If the death rate was higher than expected and the protocol considered a danger to patients they would have to disclose. Deaths due to disease natural history or those attributed to factors outside of med toxicity would likely not be disclosed. I am sure a data safety monitoring board has been revie...

Re: Can an AML study be announced before 2nd interim review?

robtmsstt1, Agree with Fish. The AML study is a preclinical study to be formally presented at a scientific meeting. It will provide some of the rationale for future clinical studies. J and J has made it clear that they will look at information from the MF/MDS clinical trials and also use this inform...

Re: Remark from CKTC on SA

Hoosier57, This is beautifully summarized by CKTC. Your second link does not work, just FYI. Thank you for sharing the post. This binary approach to apoptosis is very impressive. On another note we are now over six months from the September announcement. This means that all patients in MF study are ...

Re: New article: 'Imetelstat differentially affects normal and malignant megakaryopoiesis'

Fish, I was plenty impressed. The academic institution of origin is strong and the reaffirmation of suppression of fibrogenic Growth factors renews hope in the MF study outcome. I I think the ongoing Q2 review of patients with an otherwise expected median LE of seven months Speaks volumes. Did not i...

New article: 'Imetelstat differentially affects normal and malignant megakaryopoiesis'

The following was published on pub med and noted by muttster on YMB: Imetelstat, a telomerase inhibitor, differentially affects normal and malignant megakaryopoiesis. Pub med Mosoyan G1, Kraus T2, Ye F3, Eng K4, Crispino JD5, Hoffman R1, Iancu-Rubin C1. The importance of imetelstat suppressing fibro...

Re: MF trial study question.

I can't speak with certainty as some of the Mayo patients may have overlapped in criterion with the J and J population. Nonetheless the J and J population is distinguished by the end of the line nature of the natural history of their disease and their difficult to treat status.

Yes, they are either patients who responded to Jakafi and then stopped responding or failed to respond in the first place. In other words end stage disease with failure to respond to conventional therapy. They have about a seven month life expectancy. bp

combo therapy

delprice on YMB has posted some pretty thoughtful comments. I would love to know his scientific background. He among others has probed the possible venetoclax/imet combo for other uses. Considering Scarlett's very low key comments I think they are being very circumspect for obvious reasons. The MOLM...
