The science just changed, we didn't understand telomere replication

Just in from Rockefeller paper, everything we know is wrong. We had the very basics of DNA replication wrong. Big implications for Imetelstat. I don't see any problem for 6-thio-dG, but would like to hear any ideas on it. https://www.rockefeller.edu/news/35439-double-trouble-at-chromosome-ends/#:~:t...

The latest from MAIA-- THIO-101 works

Latest presentation from MAIA. This is bigger than PCYC... getting in at this level will let everyone recover their GERN losses and more. Happy Holidaze everyone!

Good Book On the FDA/Pharma Process

"For Blood And Money" by Vardi. Covers the insanity behind the PCYC buyout and drug approval. Apparently it helps to have a deranged Scientologist running the company... one wonders when one of our genius billionaires will realize that the FDA "system" is killing them along with us peons...

Re: ADCOM

ADCOMs do not "shorten timelines". They can kill the drug, but there is no mechanism to speed things up. That's not how FDA works.

Re: FDA accepts NDA

(?) The FDA rejected priority review for Imetelstat... EVEN THOUGH WE HAD FAST TRACK!

This is how they kill biotechs... just delay delay delay.

18,000 FDA employees, and they can't do priority review for Fast Track drug? Absurd.

Thanks for your recent posts, kmall, you make some good points. However, you can't stick with your "Bill iS a rEpTile aLiEN" stuff unless you actually believe that I have time travel tech... I have posted the subjects of op-eds and scientific papers on YHOO BEFORE THEY WERE PUBLISHED many times over...

Thanks. This poster doesn't seem to back up any SF3B1 limitations... in fact, it doesn't seem useful for further genetic targeting at all, which is unfortunate. Hopefully there is better data from the Phase 3.

Thanks for the employee list, kmall.

Imetelstat is RNA, it binds to the hTR RNA, not to proteins.

Re: Just a question

My understanding is that they shouldn't have to do a Phase 1, since Imetelstat has repeatedly been studied for safety.

As bp says, anything can be prescribed off-label (half of prescriptions are), but it's useful to have approvals in more specific indications.

Re: There are many ways to think about partnering

There is NO chance of partnering before Phase 3 results. No one dilutes their stock @$1.05 during a news drought if there were ANY chance of a partnership announcement to drive up the price and allow dilution at a better price.

Geron is going it alone at least until 2023.

The ATM Beast of Baluchistan in the room

So, the shorts think GERN is going to succeed, their covering has been the main force holding up the stock price since Dec 2020. But the CEO doesn't agree... he just keeps selling ATM shares near 52-week lows, according to the last report. If Phase 3 data is as good as Phase 2, why in the world is h...

Re: "We could have skimped and watched our pennies but....

Meanwhile, the only real research talent from the old days (Sergei Gryzanov) went to MAIA, not to Geron.

And the non-working Geron "staff" remain in Foster City. Biotechs that brag about not watching pennies, while selling ATM shares for pennies, aren't looking out for their investors.

Re: Thoughts on IMET2.0 toxicity improvements...

If they use antibody-labeled liposomes to target the oligo (e.g. the original GRN 163) on cancer cells instead of dumping most of it into off-target cells, there would clearly be a MASSIVE decrease in toxicity. But there won't be an "oral imetelstat"... MAIA's 6-thio-dG is oral, but that's because i...

Yes, they screened a lot of compounds looking for telomerase activators, and found the astragalus chemical that became TA-65. However, one million is a tiny subset of the organic chemicals, and Nature herself already has activators more powerful than TA-65, e.g. Centella asiatica herb: https://www.n...

All of which would have been VERY appropriate for Scarlett to bring up at the Investor Conference.

Both Kmall and Martin have bragged on YHOO about "reporting" any posts that contain information about Geron that they don't want others to know. Obviously Early Retirement/jkokao/Julie/etc. does the same. We really need to use other forums, YHOO's software itself deletes most posts with reference li...

Old Geron vs. New Geron

The AML combo trial is great news. News that we should have had in 2016, at least with the azacitidine arm. IMO both the Venetoclax and azacitidine arms will show massive increase in efficacy over imet monotherapy. Both because of synergy in promoting apoptosis and because it will be MUCH harder for...

rinconj, Geron was forced by the Janssen deal to shut down its lab... they don't have one. Now, repackaging the old imet into a less obsolete form for oilgo delivery is easy... but then it would have to start approval all over. There ARE many other companies using telomerase inhibitors, many combo t...
